qu’entre 60 et 65 ans [Moran, EBCC-9 2014].
Le plus grand défi posé par le traitement anti-HER2 est donc assurément celui de
l’amélioration de l’activité anti-tumorale par le double blocage (trastuzumab + autre
traitement anti-HER2 : inhibiteur tyrosine kinase ou autre anticorps anti-HER2) comme
exploré par différents programmes en néoadjuvant ou en métastatique (NeoSphere, TBCRC-
0006, NeoALTTO, Pernetta/SAKK 22/10-Unicancer UC-0140/1207/NCT01835236, EORTC
75111-10114/NCT01597414) (Gianni et al., 2012;Rimawi et al., 2013).
Ceci pourrait permettre de s’affranchir de la chimiothérapie avec sa toxicité limitante, car
même la « soft chemotherapy » dont font partie les dérivés conjugués comme le T-DM1
(Verma et al., 2012;Krop et al., 2014), peut avoir un impact important sur l’observance (Kalsi
et al., 2014).
